Abstract

This study aimed to assess the effects of hexavalent chromium on zebrafish () embryo development. The zebrafish embryos were treated with solutions containing chromium at different concentrations (0.1, 1, 3.125, 6.25, 12.5, 50, and 100 µg/mL). The development of zebrafish embryos was estimated by the determination of survival rate, heart rate, and the measurement of larvae body length. Real time RT-PCR and Western blot were performed to assess the expression of apoptosis- and antioxidant-related genes. The results showed that the reduced survival rate of zebrafish embryos and larvae was associated with an increase in chromium concentration. The exposure of higher concentrations resulted in a decrease in body length of zebrafish larvae. In addition, a marked increase in heart rate was observed in the zebrafish larvae under chromium treatment, especially at high concentrations. The real-time RT-PCR analysis showed that the transcript expressions for cell-cycle-related genes ( and ) and antioxidant-related genes ( and ) were downregulated in the zebrafish embryos treated with chromium. Western blot analysis revealed the upregulation of Caspase 3 and Bax, while a downregulation was observed in Bcl2. These results indicated that hexavalent chromium induced changes in zebrafish embryo development by altering apoptosis- and antioxidant-related genes.

本研究旨在评估六价铬对斑马鱼胚胎发育的影响。将斑马鱼胚胎用含有不同浓度(0.1、1、3.125、6.25、12.5、50和100 µg/mL)铬的溶液处理。通过测定存活率、心率以及测量幼鱼体长来评估斑马鱼胚胎的发育情况。进行实时逆转录聚合酶链反应(Real time RT-PCR)和蛋白质免疫印迹法(Western blot)以评估凋亡相关基因和抗氧化相关基因的表达。结果表明,斑马鱼胚胎和幼鱼存活率的降低与铬浓度的增加有关。较高浓度的暴露导致斑马鱼幼鱼体长减小。此外,在铬处理下的斑马鱼幼鱼中观察到心率显著增加,尤其是在高浓度时。实时逆转录聚合酶链反应分析表明,在用铬处理的斑马鱼胚胎中,细胞周期相关基因(和)以及抗氧化相关基因(和)的转录表达下调。蛋白质免疫印迹分析显示半胱天冬酶3(Caspase 3)和Bax上调,而Bcl2下调。这些结果表明,六价铬通过改变凋亡相关基因和抗氧化相关基因诱导斑马鱼胚胎发育发生变化。
