The aim of this study was to investigate the physiological response of rainbow trout () before slaughtering in the last phase of farming analyzing skin mucus and plasma. Two groups of rainbow trout were considered: Group UN ("unstressed"), represented by fish randomly captured from raceways, in the last phase of a standard fattening cycle; Group S ("stressed"), collected at the end of the pre-slaughtering tank, soon after slaughtering. The fish skin mucus was swabbed from head to tail using a sterile plastic spatula and the blood was collected through an endocardial puncture. qRT-PCR was used to study the gene expression in skin mucus. The mRNA expression levels of the IL-6 and IgD genes were higher in the S than in the Group UN. The plasma analysis showed an only a decrease in the glucose plasma levels in the Group S when compared to the Group UN. The present results indicated that the procedures adopted after slaughtering only affected changes in plasma glucose and skin mucus activity in rainbow trout suggesting that management protocol was compatible with non-stressful farming conditions.
本研究的目的是通过分析虹鳟鱼的皮肤黏液和血浆,调查养殖最后阶段宰杀前虹鳟鱼的生理反应。研究考虑了两组虹鳟鱼:UN组(“未受应激”),由标准育肥周期最后阶段从养殖池随机捕获的鱼组成;S组(“受应激”),在宰杀前水槽结束时、宰杀后不久采集。使用无菌塑料刮勺从鱼头到鱼尾擦拭鱼的皮肤黏液,并通过心内穿刺采集血液。采用qRT-PCR研究皮肤黏液中的基因表达。S组中IL-6和IgD基因的mRNA表达水平高于UN组。血浆分析显示,与UN组相比,S组血浆葡萄糖水平仅有所下降。目前的结果表明,宰杀后采用的程序仅影响虹鳟鱼血浆葡萄糖和皮肤黏液活性的变化,这表明管理方案与无应激养殖条件兼容。
