The objective of the present study was to investigate anti-inflammatory effects of disenecionyl -khellactone (DK) isolated from Thunberg, a traditional edible plant, in RAW264.7 cells stimulated with lipopolysaccharide (LPS). Anti-inflammatory effects of DK were analyzed using various techniques, including NO assay, Western blot analysis, enzyme-linked immunosorbent assay (ELISA), real-time PCR, and immunofluorescence staining. It was revealed that DK reduced the production of pro-inflammatory cytokines including Monocyte chemoattractant protein-1 (MCP-1), Tumor necrosis factor-α (TNF-α), Interleukin 1β (IL-1β), and Interleukin 6 (IL-6) in RAW264.7 cells stimulated with LPS. It was revealed that DK effectively downregulated expression levels of iNOS and COX-2 due to inhibition of NF-κB activation and suppressing the phosphorylation of p38 and jun N-terminal kinase (JNK) mitogen-activated protein kinase (MAPK) phosphorylation. Also, soluble epoxide hydrolase activity and expression were decreased by the proinflammatory inhibitor, DK. Finally, findings of this study suggest that DK isolated from might have potential as a therapeutic candidate for inflammatory diseases.
本研究的目的是调查从传统可食用植物滨蒿中分离出的去乙酰基山莴苣素(DK)对脂多糖(LPS)刺激的RAW264.7细胞的抗炎作用。使用多种技术分析了DK的抗炎作用,包括一氧化氮(NO)检测、蛋白质免疫印迹分析、酶联免疫吸附测定(ELISA)、实时聚合酶链反应(PCR)和免疫荧光染色。结果显示,DK可降低LPS刺激的RAW264.7细胞中促炎细胞因子的产生,包括单核细胞趋化蛋白-1(MCP-1)、肿瘤坏死因子-α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素6(IL-6)。结果显示,由于抑制核因子κB(NF-κB)的激活以及抑制p38和c-Jun氨基末端激酶(JNK)丝裂原活化蛋白激酶(MAPK)的磷酸化,DK有效下调了诱导型一氧化氮合酶(iNOS)和环氧化酶-2(COX-2)的表达水平。此外,促炎抑制剂DK降低了可溶性环氧化物水解酶的活性和表达。最后,本研究结果表明,从滨蒿中分离出的DK可能具有作为炎症性疾病治疗候选药物的潜力。
