s the most widely distributed and abundant species of toad in China. Bufadienolides and indole alkaloids have cardiotonic and anti-tumor activities and are important pharmacological components of . In this experiment, a novel compound () and two known compounds ( and ) were isolated and identified from the dry skin of , both of which are bufadienolides. Various column chromatography methods were used to separate and purify the extract from the dried skin of . Accurate molecular weights were measured by HR-ESI-MS, and the chemical structure of the compounds was determined by NMR spectrometers. The structures were named as (2,5,16)-2,5,16-trihydroxide bufa-14,20,22 dienolide (), gamabufotalin () and desacetylbufotalin (). cytotoxic activity assay indicated that compound showed a moderate cytotoxicity against A549 cells with IC value of 12.65 μM.
是中国分布最广、数量最多的蟾蜍种类。蟾毒配基和吲哚生物碱具有强心和抗肿瘤活性,是其重要的药理成分。在本实验中,从的干燥皮肤中分离并鉴定出一种新型化合物()和两种已知化合物(和),它们均为蟾毒配基。采用多种柱色谱方法对干燥皮肤提取物进行分离纯化。通过高分辨电喷雾电离质谱(HR-ESI-MS)测定精确分子量,并用核磁共振波谱仪确定化合物的化学结构。其结构分别命名为(2,5,16)-2,5,16-三羟基蟾毒-14,20,22-二烯醇内酯()、华蟾酥毒基()和去乙酰华蟾酥毒基()。细胞毒性活性测定表明,化合物对A549细胞表现出中等细胞毒性,IC值为12.65μM。
