Abstract

The present study aimed to figure out the potential role of exosomal microRNAs, and their targeted genes in HNC detection/diagnosis. In the present study, exosomes were extracted from the serum samples of 400 HNC patients and 400 healthy controls. Exosomes were characterized using TEM, NTA, TEM-immunogold labeling and ELISA. Quantitative PCR was used to measure the expression level of exosomal miRNA-19a, miRNA-19b and targeted genes and in HNC patients and controls. The deregulation of miR-19a (p < 0.01), miR-19b (p < 0.03), (p < 0.04) and (p < 0.04) was observed in HNC patients vs controls. ROC curve and Kaplan-Meier analysis showed the good diagnostic/prognostic value of selected exosomal microRNAs and related genes in HNC patients.

本研究旨在探讨外泌体 microRNAs 及其靶基因在头颈部癌症(HNC)检测/诊断中的潜在作用。本研究从 400 例 HNC 患者和 400 例健康对照者的血清样本中提取外泌体。使用 TEM、NTA、TEM-免疫金标记和 ELISA 对其进行鉴定。采用 qPCR 检测 HNC 患者和对照组中外泌体 miRNA-19a、miRNA-19b 及靶基因 和 的表达水平。结果显示,HNC 患者中 miR-19a(p<0.01)、miR-19b(p<0.03)、 (p<0.04)和 (p<0.04)表达下调。ROC 曲线和 Kaplan-Meier 分析表明,所选外泌体 microRNAs 和相关基因对头颈部癌症患者具有良好的诊断/预后价值。
