This study isolated a novel antioxidant peptide from black soldier fly larvae (BSFL) using enzymatic hydrolysis. Firstly, the BSFL enzymatic hydrolysate was fractionated through ultrafiltration, with the <3 kDa fraction exhibiting the strongest DPPH and ABTS radical scavenging activity. Subsequently, this fraction was further fractionated through gel filtration chromatography and RP-HPLC. Totally, 153 peptides were identified through LC-MS/MS analysis, from which a novel peptide EDEGTYKCVLS (Pep6) was screened according to activity prediction and verification. Pep6 exhibited high radical scavenging capacity and cytoprotective effect on HepG2 cells against HO damage, meanwhile significantly increasing the intracellular antioxidant enzymes activity. Molecular docking analysis indicated that Pep6 competitively bound to Keap1, thereby inhibiting the formation of Keap1-Nrf2 complex, ultimately protecting cells from oxidative stress damage. In this study, a novel antioxidant peptide Pep6 was identified from BSFL, and its antioxidant mechanism was elucidated, providing a theoretical basis for its use as a natural antioxidant.
本研究采用酶解的方法从黑水虻幼虫(BSFL)中分离得到一种新型抗氧化肽。首先,通过超滤对 BSFL 酶解产物进行分级,其中<3 kDa 级分表现出最强的 DPPH 和 ABTS 自由基清除活性。随后,该级分进一步通过凝胶过滤色谱和反相高效液相色谱进行分级。通过 LC-MS/MS 分析共鉴定出 153 种肽,根据活性预测和验证筛选出一种新型肽 EDEGTYKCVLS(Pep6)。Pep6 表现出很强的自由基清除能力和对 HepG2 细胞抵御 HO 损伤的细胞保护作用,同时显著增加细胞内抗氧化酶的活性。分子对接分析表明,Pep6 竞争性结合 Keap1,从而抑制 Keap1-Nrf2 复合物的形成,最终保护细胞免受氧化应激损伤。本研究从黑水虻中鉴定出一种新型抗氧化肽 Pep6,并阐明了其抗氧化机制,为其作为天然抗氧化剂的应用提供了理论依据。
