MicroRNAs in exosomes play a role in biological processes such as inflammation and Epithelial-mesenchymal transition (EMT). In EMT, epithelial cells undergo phenotypic changes and become similar to mesenchymal cells. EMT increases the invasion and metastasis of cancer cells. We aimed to evaluate the expression levels of miRNA-21, miRNA-218, miRNA-155, and miRNA-10b, which are effective in the pathway of inflammation and EMT in serum-derived exosome of COVID-19 patients.
Blood samples were taken from 30 patients with COVID-19 and five healthy individuals as a control group. After separating the serum from the collected blood, the exosomes were purified from the serum. Relative expression of microRNAs was measured by real-time PCR method.
The relative expression of miRNA-21, miRNA-218, and miRNA-155 in serum-derived exosomes of patients with COVID-19 had a significant increase (p < 0.0001). Also, the relative expression of miRNA-10b was significantly increased in the patient group (p < 0.01), but the changes in the expression level of miRNA-10b were not as significant as the changes in the expression level of other microRNAs.
miRNA-21, miRNA-218, miRNA-155, and miRNA-10b are involved in the pathogenesis of COVID-19 disease, and their transmission by exosomes leads to pathogenic lesions and problems in other parts of the body.
外泌体中的微小RNA在炎症和上皮-间质转化(EMT)等生物学过程中发挥作用。在EMT过程中,上皮细胞发生表型变化并变得类似于间充质细胞。EMT增加癌细胞的侵袭和转移。我们旨在评估miRNA-21、miRNA-218、miRNA-155和miRNA-10b在COVID-19患者血清来源外泌体的炎症和EMT途径中的表达水平。
采集30例COVID-19患者的血样,并选取5名健康个体作为对照组。从采集的血液中分离出血清后,从血清中纯化出外泌体。通过实时PCR法测量微小RNA的相对表达。
COVID-19患者血清来源外泌体中miRNA-21、miRNA-218和miRNA-155的相对表达显著增加(p < 0.0001)。此外,miRNA-10b在患者组中的相对表达也显著增加(p < 0.01),但miRNA-10b表达水平的变化不如其他微小RNA表达水平的变化显著。
miRNA-21、miRNA-218、miRNA-155和miRNA-10b参与COVID-19疾病的发病机制,它们通过外泌体的传递导致身体其他部位的致病损伤和问题。
