To explore the antioxidant activity of enzymatic hydrolysates of from Dalian and preliminarily elucidate their mechanisms of action both and . Samples were hydrolysed using alcalase, protamex, and neutrase. 2,2-diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging assays showed that the alcalase hydrolysate had the highest antioxidant activity, with IC values of 4.233 ± 0.067 mg/mL (DPPH) and 1.188 ± 0.066 mg/mL (ABTS). Further cell experiments indicated that the alcalase hydrolysate effectively inhibited intracellular reactive oxygen species (ROS), with a fluorescence rate of 4.29% ± 1.98%, and modulated antioxidant-related enzymes (superoxide dismutase: SOD, glutathione peroxidase: GSH-PX, malondialdehyde: MDA). The obtained optimal enzymatic hydrolysate was analysed by LC-MS/MS. The fluorescence rate of zebrafish (AAPH: 100%) decreased to 29.24% ± 1.10% (acridine orange: AO) and 56.71% ± 0.02% (ROS), with no observed embryonic toxicity. This study provides a foundation for the development of natural antioxidants.
为探究大连[具体物质未给出]酶解产物的抗氧化活性,并初步阐明其体内外作用机制。使用碱性蛋白酶、中性蛋白酶和Protamex对样品进行水解。2,2-二苯基-1-(2,4,6-三硝基苯基)肼(DPPH)和2,2'-联氮双-(3-乙基苯并噻唑啉-6-磺酸)(ABTS)自由基清除试验表明,碱性蛋白酶水解产物具有最高的抗氧化活性,DPPH法的IC值为4.233±0.067mg/mL,ABTS法的IC值为1.188±0.066mg/mL。进一步的细胞实验表明,碱性蛋白酶水解产物能有效抑制细胞内活性氧(ROS),荧光率为4.29%±1.98%,并调节抗氧化相关酶(超氧化物歧化酶:SOD、谷胱甘肽过氧化物酶:GSH-PX、丙二醛:MDA)。对获得的最佳酶解产物进行LC-MS/MS分析。斑马鱼(AAPH:100%)的荧光率降至29.24%±1.10%(吖啶橙:AO)和56.71%±0.02%(ROS),未观察到胚胎毒性。本研究为天然抗氧化剂的开发提供了基础。
