To investigate the prevalence, molecular characteristics and phylogenetic relationship of Eimeria spp. domestic alpaca at the Zhengzhou Zoo, Henan Province, China, sheather's sugar floatation method was conducted to detect coccidia in 25 fresh fecal samples. The overall prevalence of coccidia was 44.0 % (11/25), Eimeria alpacae Guerrero (1967), Eimeria lamae Guerrero (1967), and Eimeria punoensis Guerrero (1967) were identified by their morphological characteristics. The 18S rRNA and COI loci sequences were sequenced for molecular biological identification, homology comparison, and phylogenetic analysis of Eimeria spp. by single oocyst selection technology and multi-locus nested PCR amplification. The sequence homology among the three Eimeria spp. from alpaca at the 18S rRNA and COI gene loci were 98.8-99.4 % and 94.6-97.7 %, respectively. At the 18S rRNA and COI loci, the three Eimeria spp. exhibited high identity with Eimeria spp. from goats and sheep. To our knowledge, this is the first report on the molecular identification and evolutionary analysis of three Eimeria spp. from alpaca. This study also provides a more accurate identification method for Eimeria spp. from alpaca.
为调查中国河南省郑州市动物园圈养羊驼艾美耳球虫的感染率、分子特征及系统发育关系,采用Sheather氏糖漂浮法对25份新鲜粪便样本进行球虫检测。球虫总感染率为44.0%(11/25),通过形态学特征鉴定出阿尔帕卡艾美耳球虫(Eimeria alpacae Guerrero,1967)、拉玛艾美耳球虫(Eimeria lamae Guerrero,1967)和普诺艾美耳球虫(Eimeria punoensis Guerrero,1967)。采用单卵囊挑选技术和多位点巢式PCR扩增,对艾美耳球虫的18S rRNA和COI基因座序列进行测序,用于分子生物学鉴定、同源性比较和系统发育分析。来自羊驼的三种艾美耳球虫在18S rRNA和COI基因座的序列同源性分别为98.8 - 99.4%和94.6 - 97.7%。在18S rRNA和COI基因座上,这三种艾美耳球虫与来自山羊和绵羊的艾美耳球虫具有高度同一性。据我们所知,这是关于羊驼三种艾美耳球虫分子鉴定和进化分析的首次报道。本研究还为羊驼艾美耳球虫提供了一种更准确的鉴定方法。
