Abstract

ETHNOPHARMACOLOGICAL RELEVANCE

Citri Sarcodactylis Fructus (CSF), a Chinese herbal medicine, is recorded to exert the effect of drying dampness and eliminating phlegm. It is often used for the treatment of coughs with excessive phlegm. However, the material basis and mechanism of the expectorant effect of CSF remain unclear.

AIM OF THE STUDY

This study aims to investigate the active substance basis, molecular mechanism, and blood exposure effect of CSF.

METHODS

The effective component group of CSF was identified through spectrum-effect relationship analysis. Metabolic biomarkers were identified through metabolomic analysis, and metabolic pathways were analyzed. Network pharmacology and molecular docking were used to predict relevant targets and signal pathways, and western blotting was used to verify the expression levels of the targets. Pharmacokinetic analysis was performed to clarify the changes in the contents of representative components of CSF in blood.

RESULTS

Pharmacodynamic results showed that the 70 % ethanol elution fraction (70 % EF) of CSF had a good expectorant effect. Combined with spectrum-effect relationship and network pharmacology analysis, six main active components were screened out, including scoparone, 5,6,7-trimethoxycoumarin, 5,7,3'-trihydroxy-6,4',5'-trimethoxyflavone, 5,7-dimethoxycoumarin, limonin, and nomilin. Metabolomic analysis revealed that the expectorant effect of CSF may be relevant to phenylalanine metabolism; glycerol phospholipid metabolism; and phenylalanine, tyrosine, and tryptophan biosynthesis. Based on the western blotting analysis, CSF might exert its expectorant function via targeting AKT and EGFR. The results of pharmacokinetic analysis revealed that all the eight compounds (isorhamnetin, diosmetin, 5,7,3'-trihydroxy-6,4',5'-trimethoxyflavone, 5,7-dimethoxycoumarin, bergaptol, bergapten, oxypeucedanin, and byakangelicol) could be effectively absorbed in rat plasma and had good bioavailability.

CONCLUSION

The 70 % EF of CSF had a good expectorant effect. The material basis and mechanism of the expectorant effect of CSF were preliminarily revealed by combining multiple analyses.
