Abstract

Depending on its severity, spinal cord injury (SCI) can cause bowel and bladder incontinence, sensory abnormalities, muscle paralysis, and lifelong disability. Enhancing the biological properties of stem cells before transplantation can improve the efficacy of cell therapy. Resveratrol has recently been used to enhance mesenchymal stem cells' (MSCs') paracrine capacity for tissue repair. Our goal was to implant mesenchymal stem cells (MSCs) into a rat spinal cord injury (SCI) model after pre-treating them with resveratrol. First, MSCs were isolated from adipose tissue. Then, the cells were treated with resveratrol, and the amount of nerve growth factor (NGF) in the supernatant was measured. Rats were randomly assigned to four groups and given a weight-drop technique to cause spinal cord injury. MSCs were injected into the site of injury 30 min after SCI. A functional recovery test was conducted over four weeks. Luxal fast blue and hematoxylin-eosin staining were done to evaluate the tissues. The concentration of 0.2 micromolar of resveratrol increased the expression of NGF in the supernatant of MSCs. There was a significant increase in the average motor function scores in the fourth week after the injury in animals that received stem cells treated with resveratrol compared to the SCI group. This study demonstrated that transplanted MSCs treated with resveratrol exhibit better efficacy in promoting functional recovery.
