Abstract

BACKGROUND

Spexin is an adipokine secreted by various tissues, including white adipocytes and the brain. Its release is inversely associated with the state of energy storage. Curcumin (CUR), a polyphenolic substance extracted from turmeric, has been widely investigated for its beneficial effects, including the inhibition of oxidative stress and inflammation. Nano-curcumin (nCUR) is a novel preparation developed to enhance curcumin's absorption and therapeutic potency. This present study aimed to examine the comparative impacts of CUR and nCUR on plasma levels of Spexin, its gene expression in liver tissue, insulin resistance, lipid profiles, and fasting blood glucose (FBG) in type 2 diabetes mellitus (T2DM) rats.

METHODS

Twenty-four male Wistar rats were divided into four groups: A T2DM group receiving 200 mg/kg nCUR, a T2DM group receiving 200 mg/kg CUR, a T2DM group, and a control group. The rats in the T2DM groups developed diabetes through a high-fat and high-glucose diet, along with streptozotocin injection. Plasma levels of Spexin, FBG, insulin resistance, and lipid profile were assessed. Additionally, the Spexin gene expression in liver samples was evaluated using quantitative real-time PCR (qRT-PCR).

RESULTS

Both nCUR and CUR significantly improved insulin resistance, reduced FBG, and improved lipid levels in T2DM rats compared to the diabetic control rats (p < 0.05). Notably, nCUR demonstrated a more pronounced elevation in plasma Spexin levels and its gene expression in liver tissue than CUR.

CONCLUSION

nCUR offers superior therapeutic benefits over CUR in managing T2DM, particularly in improving lipid metabolism, and Spexin regulation.

背景

Spexin是一种由包括白色脂肪细胞和大脑在内的多种组织分泌的脂肪因子。其释放与能量储存状态呈负相关。姜黄素(CUR)是从姜黄中提取的一种多酚类物质,因其有益作用,包括抑制氧化应激和炎症,已得到广泛研究。纳米姜黄素(nCUR)是为提高姜黄素的吸收和治疗效果而开发的一种新型制剂。本研究旨在探讨CUR和nCUR对2型糖尿病(T2DM)大鼠血浆Spexin水平、其在肝组织中的基因表达、胰岛素抵抗、血脂谱和空腹血糖(FBG)的比较影响。

方法

将24只雄性Wistar大鼠分为四组:一组T2DM大鼠接受200mg/kg nCUR,一组T2DM大鼠接受200mg/kg CUR,一组T2DM大鼠,以及一组对照组。T2DM组大鼠通过高脂高糖饮食联合链脲佐菌素注射诱导糖尿病。评估血浆Spexin水平、FBG、胰岛素抵抗和血脂谱。此外,使用定量实时PCR(qRT-PCR)评估肝组织样本中Spexin基因的表达。

结果

与糖尿病对照组大鼠相比,nCUR和CUR均显著改善了T2DM大鼠的胰岛素抵抗,降低了FBG,并改善了血脂水平(p<0.05)。值得注意的是,nCUR在提高血浆Spexin水平及其在肝组织中的基因表达方面比CUR表现得更为明显。

结论

在管理T2DM方面,nCUR比CUR具有更好的治疗效果,特别是在改善脂质代谢和Spexin调节方面。
