Abstract

Rosette inhibition (RI) was obtained in the absence of complement, but was generally much enhanced by the presence of complement. RI titres measured in the presence of complement correlated well with immunosuppressive potency. In the absence of complement, RI was due to anti-immunoglobulin antibodies contained in ALS and did not correlate with immunosuppressive potency. Complement enhancement of RI required only C[unk]1 and C4 and C[unk]1 used alone had a significant enhancing potency. The effect of complement developed within 5 minutes on ALS-treated lymphocytes. Rosette inhibition was obtained with concentrations of ALS and complement much lower than those necessary for cytotoxicity. With crude rabbit ALS a competition was observed between inactivated rabbit complement and guinea-pig complement. Lymphocytotoxicity was obtained after 90 minutes' incubation of lymphocytes with ALS in the presence of complement. Rabbit ALS was cytotoxic in the presence of rabbit or guinea-pig complement. Horse ALS fixed rabbit but not guinea-pig complement. The whole sequence of the nine components was necessary for the cytotoxic reaction.
