The effect of whole body γ irradiation of donors on the cytotoxic action of spleen cells from specifically sensitized rats towards the xenogeneic Chang cell was studied. The target cells in this model are killed by effector lymphocytes which are triggered into cytotoxic activity by IgG complexed with target cell antigens. A separate population of lymphocytes produces the sensitizing antibody. It was shown that production of sensitizing antibody could not be induced within 16 days after 850 rad whole body irradiation. The cytotoxic effector population, however, despite initial severe depletion, neared complete recovery by this time. The implications of this differential effect are discussed.
