It has long been known that testes arise only in the presence of a Y chromosome. The existence of a hypothetical gene, the testis-determining factor (TDF) on the Y chromosome, has therefore been postulated. SRY is the gene located in the sex-determining region of the human Y chromosome and has many of the properties expected for TDF. To evaluate the value of SRY PCR in sex determination, individuals with ambiguous genitalia were selected for study. By identification of the DNA sequence of SRY, a 609-bp fragment of PCR product, sex determination could be successfully made.
长期以来,人们一直知道睾丸仅在存在Y染色体的情况下才会出现。因此,有人推测Y染色体上存在一种假设的基因——睾丸决定因子(TDF)。SRY是位于人类Y染色体性别决定区域的基因,具有TDF所预期的许多特性。为了评估SRY聚合酶链反应(PCR)在性别鉴定中的价值,选择了生殖器模糊的个体进行研究。通过鉴定SRY的DNA序列,即PCR产物的一个609碱基对片段,可以成功进行性别鉴定。
