BACKGROUND/AIMS: The aim of this study was to try to understand the effects of the synthetic matrix metalloproteinase inhibitor Batimastat (BB-94) on hepatocellular carcinoma (HCC).
An orthotopic metastatic human hepatocellular carcinoma in nude mice model (LCI-D20) was used to study primary tumor growth, local invasion and metastasis of HCC. MTT assay was used to study the effects of BB-94 on cytotoxin and proliferation of HCC cell line SMMC-7721 in vitro. A gelatine zymograph was used to study the expression of MMPs in the LCI-D20 tumor tissue.
BB-94 can inhibit primary tumor growth, local invasion, intrahepatic and lung metastasis, as well as prolong survival. BB-94 did not affect the proliferation of HCC cells in vitro. LCI-D20 tumor tissue expresses MMP-2 and MMP-9.
BB-94 has a cytostatic therapeutic effect on HCC.
背景/目的:本研究旨在了解合成基质金属蛋白酶抑制剂batimastat(BB - 94)对肝细胞癌(HCC)的影响。
采用裸鼠原位转移性人肝细胞癌模型(LCI - D20)研究HCC的原发性肿瘤生长、局部侵袭和转移。采用MTT法研究BB - 94对肝癌细胞系SMMC - 7721体外细胞毒性和增殖的影响。用明胶酶谱法研究LCI - D20肿瘤组织中基质金属蛋白酶(MMPs)的表达。
BB - 94可抑制原发性肿瘤生长、局部侵袭、肝内和肺转移,并延长生存期。BB - 94在体外不影响肝癌细胞的增殖。LCI - D20肿瘤组织表达MMP - 2和MMP - 9。
BB - 94对HCC具有细胞生长抑制治疗作用。
