To study the effects of huperzine-A on neuromuscular junction transmission in mouse.
The isolated mouse phrenic nervehemidiaphragm preparations were used with the conventional intracellular recording technique. The spontaneous electrical activities of cholinergic nerve terminals (miniature end-plate potentials, MEPP) were recorded.
Huperzine-A, tacrine, and E2020 at the concentrations of 0.05-1 mumol.L-1 increased the amplitude, mean rise time, and half decay time of MEPP in a concentration-dependent manner. Their potencies were E2020 > huperzine-A > tacrine.
The anticholinesterase action of huperzine-A in cholinergic synapses is stronger than that of tacrine.
研究石杉碱甲对小鼠神经肌肉接头传递的影响。
采用常规细胞内记录技术,制备离体小鼠膈神经 - 半膈肌标本。记录胆碱能神经末梢的自发电活动(微小终板电位,MEPP)。
浓度为0.05 - 1 μmol·L-1的石杉碱甲、他克林和E2020均以浓度依赖的方式增加MEPP的幅度、平均上升时间和半衰减时间。它们的效能为E2020 > 石杉碱甲 > 他克林。
石杉碱甲在胆碱能突触中的抗胆碱酯酶作用强于他克林。
