The aim of this study on the genetic polymorphism at FES locus was to know whether there is genetic relationship between Chinese and German populations.
EDTA-blood specimens were collected from 311 healthy unrelated Han individuals in Jilin, Chengdu and Guangzhou in China and from 123 healthy unrelated individuals in Germany. The DNA samples were extracted using Chelex method and amplified by PCR technique. The PAGE horizontal electrophoresis was used for typing the PCR product.DNA sequences were analyzed by ALF.
There were nine alleles at FES locus in the Chinese population. The allele frequencies were FES6,0-0. 0051; FES 8,0-0.0042; FES9,0.0083-0.0202; FES10,0.0202-0.0604; FES11,0.4066-0.5101; FES12,0.2424-0.3099;FES13, 0.1860-0.2198; FES14,0.0041-0.0165; and FES15,0-0.0050, respectively. In the German population, a variation in 5 flanking region and nine alleles at FES locus were noted. The allele frequencies were FES8,0.0120; FES10a,0.02317; FES10,0.0407; FES11a,0.0122; FES11,0.4146; FES12a, 0.0041; FES12, 0.2357; FES13, 0.0447; and FES14, 0.0041, respectively. The results of test for Hardy-Weinberg equilibrium showed that the genotype distributions observed in the populations were correspondent with the expected.
There is a remarkable difference in the distribution of allele frequencies at FES locus between Chinese and German populations.
本研究对FES基因座的遗传多态性进行分析,旨在了解中国人群与德国人群之间是否存在遗传关系。
采集了来自中国吉林、成都和广州的311名健康无血缘关系的汉族个体以及来自德国的123名健康无血缘关系个体的乙二胺四乙酸抗凝血标本。采用Chelex法提取DNA样本,并通过聚合酶链反应(PCR)技术进行扩增。采用聚丙烯酰胺凝胶水平电泳对PCR产物进行分型。通过自动激光荧光测序仪(ALF)分析DNA序列。
中国人群FES基因座有9个等位基因。其等位基因频率分别为:FES6,0 - 0.0051;FES8,0 - 0.0042;FES9,0.0083 - 0.0202;FES10,0.0202 - 0.0604;FES11,0.4066 - 0.5101;FES12,0.2424 - 0.3099;FES13,0.1860 - 0.2198;FES14,0.0041 - 0.0165;FES15,0 - 0.0050。在德国人群中,发现FES基因座5侧翼区存在变异且有9个等位基因。其等位基因频率分别为:FES8,0.0120;FES10a,0.02317;FES10,0.0407;FES11a,0.0122;FES11,0.4146;FES12a,0.0041;FES12,0.2357;FES13,0.0447;FES14,0.0041。哈迪-温伯格平衡检验结果表明,观察到的人群基因型分布与预期相符。
中国人群和德国人群在FES基因座的等位基因频率分布上存在显著差异。
