To study the role of mast cells in hypertensive myocardial fibrosis.
The myocardial collagen and the degree of myocardial fibrosis were assessed using Masson's trichrome staining and image analysis plus toluidine blue as well as proliferating cell nuclear antigen (PCNA) staining and transmission electronic microscopy for SHRsp, aged 34 weeks (S group, n = 6). Besides, age and sex-matched WKY (W group, n = 5) were used as the controls.
MC count increased 3.9 folds and PCNA immunostaining positive cells (PCNAP) increased 3.9 fold (both P < 0.0001) in S vs W group. MC were positively correlated with the interstitial collagen volume fraction (ICVF), perivascular collagen area/vascular lumenal cross area (PVCA/VLCA), and PCNAP (r = 0.47, P < 0.05; 0.59, P < 0.001; 0.68, P < 0.001 respectively). Electromicroscopy showed specific piecemeal degranulation of the mast cells, locating between the praliferative interstitial collagen fibers.
The findings suggested that MCs play a role in certain degree to the myocardial fibrosis present in SHRsp.
研究肥大细胞在高血压性心肌纤维化中的作用。
采用Masson三色染色和图像分析以及甲苯胺蓝对34周龄的自发性高血压大鼠(SHRsp,S组,n = 6)评估心肌胶原和心肌纤维化程度,同时采用增殖细胞核抗原(PCNA)染色和透射电子显微镜观察。此外,选取年龄和性别匹配的Wistar-Kyoto大鼠(WKY,W组,n = 5)作为对照。
与W组相比,S组肥大细胞计数增加3.9倍,PCNA免疫染色阳性细胞(PCNAP)增加3.9倍(均P < 0.0001)。肥大细胞与间质胶原容积分数(ICVF)、血管周围胶原面积/血管腔横截面积(PVCA/VLCA)和PCNAP呈正相关(r分别为0.47,P < 0.05;0.59,P < 0.001;0.68,P < 0.001)。电镜显示肥大细胞有特异性的颗粒逐片脱颗粒现象,位于增殖的间质胶原纤维之间。
研究结果提示肥大细胞在一定程度上参与了SHRsp的心肌纤维化过程。
