Abstract

OBJECTIVE

To study the protective effect of removing circulatory TNF by immunoadsorption on endotoxin-induced acute lung injury in rabbits.

METHOD

New Zealand white rabbits injected with lethal dose of endotoxin underwent hemoperfusion with immunoadsorbent. TNF and IL-8 levels in BALF were measured, the number of leukocytes and percentage of PMN in BALF were analyzed and the ratio of wet lung to body weight and pulmonary microvascular permeability index were also studied. In addition, the survival rate was observed.

RESULT

Treatment with immunoadsorption markedly attenuated the pulmonary microvascular permeability increased by endotoxin and improved the survival rate.

CONCLUSION

Immunoadsorption to remove circulating TNF is an effective way to protect rabbits against endotoxin-induced acute lung injury.

目的

研究通过免疫吸附去除循环中的肿瘤坏死因子(TNF)对家兔内毒素诱导的急性肺损伤的保护作用。

方法

对注射致死剂量内毒素的新西兰白兔进行免疫吸附剂血液灌流。检测支气管肺泡灌洗液(BALF)中TNF和白细胞介素-8(IL-8)水平,分析BALF中白细胞数量和中性粒细胞百分比,同时研究肺湿重与体重之比及肺微血管通透性指数。此外,观察生存率。

结果

免疫吸附治疗显著减轻了内毒素所致的肺微血管通透性增加,并提高了生存率。

结论

免疫吸附去除循环中的TNF是保护家兔免受内毒素诱导的急性肺损伤的有效方法。
