To investigate FasL expression in hilar cholangiocarcinoma tissues and cultured cholangiocarcinoma cells, and to assess its ability to induce apoptosis.
We studied the expression of FasL by human hilar cholangiocaroinomas tissues by immunohistochemistry, and the QBC939 cholangiocarcinoma cell line by RT-PCR, immunohistochemistry, and Western Blot. TUNEL and flow cytometry were used to detect apoptotic cells.
Prevalent expression of FasL was detected in 39 resected hilar cholangiocarcinoma tissues. TUNEL staining disclosed a high level of cell death among lymphocytes infiltrating FasL positive areas of tumor. FasL mRNA and protein expressions in cholangiocarcinoma cells could induce Jurkat cells.
Hilar cholangiocarcinomas may elude immunological surveillance by inducing, via Fas/FasL system, the apoptosis of activated lymphocytes.
研究FasL在肝门部胆管癌组织及培养的胆管癌细胞中的表达,并评估其诱导细胞凋亡的能力。
我们采用免疫组织化学方法研究人肝门部胆管癌组织中FasL的表达,采用逆转录聚合酶链反应(RT-PCR)、免疫组织化学和蛋白质印迹法研究QBC939胆管癌细胞系中FasL的表达。采用末端脱氧核苷酸转移酶介导的缺口末端标记法(TUNEL)和流式细胞术检测凋亡细胞。
在39例切除的肝门部胆管癌组织中检测到FasL的普遍表达。TUNEL染色显示,在肿瘤FasL阳性区域浸润的淋巴细胞中细胞死亡水平较高。胆管癌细胞中的FasL mRNA和蛋白表达可诱导Jurkat细胞凋亡。
肝门部胆管癌可能通过Fas/FasL系统诱导活化淋巴细胞凋亡,从而逃避免疫监视。
