Abstract

OBJECTIVE

To investigate the relationship between the haze formation and the change of extracellular matrix (ECM) in the corneal tissue after photorefractive keratectomy (PRK).

METHODS

Twenty-four adult New Zealand White rabbits received PRK in the right eye for minus sign10.00 D ablation. According to the time of rabbits sacrifice, they were divided into 8 groups, and 3 rabbits were in each group. They were immediate, 24 hour, 1 week, 2 week, 1 month, 3 month, 6 month and 12 month groups. After surgery, the corneas were examined periodically by slip-lamp microscopy for haze formation. The right eyes were enucleated, and the corneas were studied with immunohistochemical evaluation for extracellular matrix, including collagen type I, type III, type VI, cellular fibronectin, tenascin and laminin.

RESULTS

The incidence of haze was 100% after PRK. The formation of haze had a tendency from mild to severe, and along with the time prolongation, it gradually decreased and disappeared. The newly synthesized collagen type I, type III, type VI, cellular fibronectin, tenascin and laminin were deposited in the anterior corneal stroma.

CONCLUSION

The formation of haze is closely related to the deposition of ECM elements in the anterior corneal stroma in the surgical area.

目的

探讨准分子激光原位角膜磨镶术(PRK)后角膜组织中 haze 形成与细胞外基质(ECM)变化之间的关系。

方法

24 只成年新西兰白兔右眼接受 -10.00 D 的 PRK 切削。根据兔子处死时间分为 8 组,每组 3 只。分别为即刻、24 小时、1 周、2 周、1 个月、3 个月、6 个月和 12 个月组。术后定期用裂隙灯显微镜检查角膜 haze 形成情况。摘除右眼,对角膜进行免疫组织化学评估以检测细胞外基质,包括Ⅰ型、Ⅲ型、Ⅵ型胶原、细胞纤连蛋白、腱生蛋白和层粘连蛋白。

结果

PRK 后 haze 发生率为 100%。haze 形成有从轻度到重度的趋势,并随时间延长逐渐减轻直至消失。新合成的Ⅰ型、Ⅲ型、Ⅵ型胶原、细胞纤连蛋白、腱生蛋白和层粘连蛋白沉积于角膜前基质层。

结论

haze 的形成与手术区域角膜前基质层中 ECM 成分的沉积密切相关。
