To evaluate the effect of human telomerase reverse transcriptase (hTERT) gene antisense oligodeoxynucleotide (As-ODN) on telomerase activity and cell apoptosis in colon cancer cell line SW480.
As-ODN was transfected into cells SW480 by liposomal transfection. Cultured cells were divided into three groups: ASODN (5'GGAGCGCGCGGCATCGCGGG-3'), sense oligodeoxynucleotide (5'-CCCGCGATGCCGCGCGCTCC-3', S-ODN) and control. The concentration of oligodeoxynucleotide and liposome was 10 micromol/L and 16 mg/L, respectively. The activity of telomerase was examined by telomeric repeat amplification protocol (TRAP)-enzyme-linked immunosorbent assay (ELISA), and cell apoptosis was observed by morphology and flow cytometry in each group.
Telomerase activity began to be down-regulated or inhibited when cells SW480 were treated with As-ODN for 72 h, and cell apoptosis was induced.
It is suggested that hTERT As-ODN might specially inhibit the activity of telomerase in colon cancer cells and it is further proved that the hTERT gene has a significant correlation with telomerase activity. Further evidence is needed to prove whether hTERT As-ODN is a potential tool for the treatment of colon cancer.
评估人端粒酶逆转录酶(hTERT)基因反义寡脱氧核苷酸(As-ODN)对结肠癌细胞系SW480中端粒酶活性及细胞凋亡的影响。
采用脂质体转染法将As-ODN转染至SW480细胞。将培养的细胞分为三组:ASODN(5'GGAGCGCGCGGCATCGCGGG-3')、正义寡脱氧核苷酸(5'-CCCGCGATGCCGCGCGCTCC-3',S-ODN)和对照组。寡脱氧核苷酸和脂质体的浓度分别为10微摩尔/升和16毫克/升。采用端粒重复序列扩增法(TRAP)-酶联免疫吸附测定(ELISA)检测端粒酶活性,通过形态学和流式细胞术观察每组细胞凋亡情况。
用As-ODN处理SW480细胞72小时后,端粒酶活性开始下调或受到抑制,并诱导细胞凋亡。
提示hTERT As-ODN可能特异性抑制结肠癌细胞中端粒酶的活性,进一步证明hTERT基因与端粒酶活性显著相关。hTERT As-ODN是否为治疗结肠癌的潜在工具还需进一步证据证实。
