Abstract

The overall objective of this study was to determine the effect of gossypol on human lymphocytes. Blood samples were taken from healthy donors and lymphocytes were cultured with various concentrations of gossypol (25-150 microM). The percentage of apoptotic cells was determined by assessing DNA fragmentation by agarose gel electrophoresis; morphological features of apoptosis were assessed by light microscopy. The concentrations of gossypol required to induce apoptosis in human lymphocytes without causing necrosis through cytotoxic effects were between 25-50 microM.

本研究的总体目标是确定棉酚对人淋巴细胞的影响。从健康供体采集血样,将淋巴细胞与不同浓度的棉酚(25 - 150微摩尔)进行培养。通过琼脂糖凝胶电泳评估DNA片段化来确定凋亡细胞的百分比;通过光学显微镜评估凋亡的形态学特征。在不通过细胞毒性作用引起坏死的情况下,诱导人淋巴细胞凋亡所需的棉酚浓度在25 - 50微摩尔之间。
