To investigate the proliferation and plasticity of neural stem cells in situ in adult rats after cerebral infarction.
Cerebral infarction models of rats were made and the dynamic expression of bromodeoxyuridine (BrdU) and BrdU/polysialylated neural cell adhesion molecule (PSA-NCAM) were determined by immunohistochemistry and immunofluorescence staining.
Compared with the controls, the number of BrdU-positive cells in the subventricular zone (SVZ) and hippocampus increased strikingly at day 1 (P < 0.05), reached maximum at day 7, and decreased markedly at day 14, but it was still elevated compared with that of the controls (P < 0.05); The number of BrdU-labeled with PSA-NCAM-positive cells increased strikingly at day 7 (P < 0.05), reached maximum at day 14, and markedly decreased at day 28, but it was still elevated compared with that of the controls (P < 0.05), and was equal to 60% of the number of BrdU-positive cells in the same period.
Our results indicate that cerebral infarction stimulate the proliferation of inherent neural stem cells in situ and most proliferated neural stem cells represent neural plasticity.
研究成年大鼠脑梗死灶内神经干细胞的增殖及可塑性。
制作大鼠脑梗死模型,采用免疫组织化学和免疫荧光染色法检测溴脱氧尿苷(BrdU)及BrdU/多唾液酸神经细胞黏附分子(PSA-NCAM)的动态表达。
与对照组相比,梗死灶周围室管膜下区(SVZ)和海马区BrdU阳性细胞数在第1天显著增加(P<0.05),第7天达到高峰,第14天明显下降,但仍高于对照组(P<0.05);BrdU标记的PSA-NCAM阳性细胞数在第7天显著增加(P<0.05),第14天达到高峰,第28天明显下降,但仍高于对照组(P<0.05),且为同期BrdU阳性细胞数的60%。
结果表明,脑梗死可刺激内源性神经干细胞原位增殖,且大部分增殖的神经干细胞具有神经可塑性。
