To study the effect of parthenolide on the proliferation of vascular smooth muscle cell(VSMC) and its mechanism.
Vascular smooth muscle cell was cultured, the protein levels of c-fos, c-myc, p15, p16, p18, p19 were measured by Western blot method, cell cycle were examined with flow cytometry, and the DNA synthesis was determined by [3H]-TdR incorporation.
Parthenolide inhibited protein levels of c-fos, c-myc in a time-dependent manner but didn't affect the protein levels of p15, p16, p18, p19. Flow cytometric DNA analysis revealed that parthenolide increased significantly G0/G1 phase of VSMC and decreased S phase of VSMC in a dose-dependent manner. Parthenolide inhibited [3H]-TdR incorporation in a dose dependent manner.
Parthenolide may inhibit proliferation of VSMC by inhibiting the expressions of c-fos, c-myc, but not the expressions of p15, p16, p18, p19.
研究小白菊内酯对血管平滑肌细胞(VSMC)增殖的影响及其机制。
培养血管平滑肌细胞,采用蛋白质免疫印迹法检测c-fos、c-myc、p15、p16、p18、p19的蛋白水平,用流式细胞术检测细胞周期,通过[3H]-TdR掺入法测定DNA合成。
小白菊内酯能时间依赖性地抑制c-fos、c-myc的蛋白水平,但不影响p15、p16、p18、p19的蛋白水平。流式细胞仪DNA分析显示,小白菊内酯能剂量依赖性地使VSMC的G0/G1期显著增加,S期减少。小白菊内酯能剂量依赖性地抑制[3H]-TdR掺入。
小白菊内酯可能通过抑制c-fos、c-myc的表达,而不是p15、p16、p18、p19的表达来抑制VSMC的增殖。
