Abstract

OBJECTIVE

To investigate the MOST-1 mRNA expression in bone marrow (BM) mononuclear cells in children with acute lymphoblastic leukemia, and to explore its association with immunophenotype and treatment response.

METHODS

Semiquantitative RT-PCR was employed to study the MOST-1 mRNA expression in BM mononuclear cells separated by Ficoll density gradient method. The MOST-1 expression levels were represented by the ratio of MOST-1 band pixels against its corresponding housekeeping gene beta-actin mRNA band pixels determined by GDS8000 densitometry and GelWork-1 analysis software. The PCR product was eluted and sequenced, and its sequence was confirmed by Pairwise BLAST search.

RESULTS

A total of 17 children with acute lymphoblastic leukemia were studied. MOST-1 mRNA was exclusively expressed in the mononuclear cells from 3 patients with ALL-L3 type. However, there was no MOST-1 mRNA expression in other 14 children with ALL-L1 or ALL-L2, irrespective of their initial peripheral WBC count and blast cell percentage in peripheral blood and bone marrow. The MOST-1 mRNA expression levels in two of the ALL-L3 patients with higher blast cell percentages in peripheral blood and bone marrow were 3- and 2-fold respectively as compared with that in the third ALL-L3 child with lower initial blast cell load. MOST-1 mRNA expression was no longer detected in the two ALL-L3 children after complete remission with combination chemotherapy.

CONCLUSION

MOST-1 was expressed in the bone marrow mononuclear cells in patients with ALL-L3, and its expression level was somewhat related to tumor cell burden. It might be implicated in the leukemogenesis of ALL-L3 and might serve as an indicator of tumor burden and thus a useful guide for clinical management.

目的

研究急性淋巴细胞白血病患儿骨髓单个核细胞中MOST-1 mRNA的表达情况,并探讨其与免疫表型及治疗反应的关系。

方法

采用半定量逆转录聚合酶链反应(RT-PCR)检测经Ficoll密度梯度离心法分离的骨髓单个核细胞中MOST-1 mRNA的表达。MOST-1的表达水平通过GDS8000密度测定仪和GelWork-1分析软件测定MOST-1条带像素与其相应看家基因β-肌动蛋白mRNA条带像素的比值来表示。PCR产物经洗脱和测序,其序列通过成对BLAST搜索进行确认。

结果

共研究了17例急性淋巴细胞白血病患儿。MOST-1 mRNA仅在3例ALL-L3型患者的单个核细胞中表达。然而,其他14例ALL-L1或ALL-L2患儿无论其外周血白细胞初始计数及外周血和骨髓中原始细胞百分比如何,均未检测到MOST-1 mRNA表达。外周血和骨髓中原始细胞百分比较高的2例ALL-L3患者的MOST-1 mRNA表达水平分别是初始原始细胞负荷较低的第3例ALL-L3患儿的3倍和2倍。联合化疗完全缓解后,2例ALL-L3患儿未再检测到MOST-1 mRNA表达。

结论

MOST-1在ALL-L3患者的骨髓单个核细胞中表达,其表达水平与肿瘤细胞负荷有一定关系。它可能参与ALL-L3的白血病发生过程,可能作为肿瘤负荷的指标,从而为临床治疗提供有用的指导。
