To explore the immune response of mice vaccinated with HIV-2 gp105 nucleic acid vaccine.
pVAX1-gp105 recombinant DNA vaccine was constructed by inserting HIV-2 gp105 cDNA into an eukaryotic expression vector pVAX1. BALB/c mice were immunized with pVAX1-gp105, pVAX1 and PBS, respectively via intramuscular injection. Anti-HIV-2 antibody was detected by ELISA. The percentages of CD4(+) and CD8(+) T cell subsets from immunized mice were analyzed by flow cytometry and the specific killing activity of splenic CTLs was measured by lactate dehydrogenase (LDH) release assay.
The titer of specific antibody, the numbers of CD4(+) and CD8(+) T cells and cytotoxic activity of specific CTLs in pVAX1-gp105 vaccination group were obviously higher than those in control group (P<0.01, P<0.05 and P<0.01, respectively).
pVAX1-gp105 nucleic acid vaccine can elicit specific cellular and humoral immunities in mice.
探讨用HIV-2 gp105核酸疫苗免疫小鼠后的免疫反应。
将HIV-2 gp105 cDNA插入真核表达载体pVAX1构建pVAX1-gp105重组DNA疫苗。通过肌肉注射分别用pVAX1-gp105、pVAX1和PBS免疫BALB/c小鼠。用ELISA检测抗HIV-2抗体。用流式细胞术分析免疫小鼠的CD4(+)和CD8(+) T细胞亚群百分比,并用乳酸脱氢酶(LDH)释放试验检测脾细胞CTL的特异性杀伤活性。
pVAX1-gp105疫苗接种组的特异性抗体滴度、CD4(+)和CD8(+) T细胞数量以及特异性CTL的细胞毒活性均明显高于对照组(分别为P<0.01、P<0.05和P<0.01)。
pVAX1-gp105核酸疫苗可在小鼠中引发特异性细胞免疫和体液免疫。
