Abstract

AIM

To study the function of alpha-fetoprotein (AFP) in SMMC-7721 hepatoma cells.

METHODS

A hairpin siRNA expressing plasmid pSilencer3.0-H1-afp was constructed and transfected into SMMC-7721 cells with Lipofectamine 2000. The expression of AFP was monitored by real-time RT-PCR and immunoassays, its effect on SMMC-7721 cell proliferation and cell death was detected by MTT and fluorescence activated cell sorter (FACS).

RESULTS

The AFP-siRNA expressing plasmid downregulated the expression of AFP obviously (about 34%), and inhibited SMMC-7721 cell proliferation, but did not induce apoptosis.

CONCLUSION

Downregulation of AFP siRNA inhibits proliferation of SMMC-7721 cells, but cannot cause apoptosis.

目的

研究甲胎蛋白(AFP)在SMMC - 7721肝癌细胞中的功能。

方法

构建表达短发夹RNA的质粒pSilencer3.0 - H1 - afp,并使用脂质体2000将其转染至SMMC - 7721细胞中。通过实时逆转录聚合酶链反应(RT - PCR)和免疫测定法监测AFP的表达,采用MTT法和荧光激活细胞分选仪(FACS)检测其对SMMC - 7721细胞增殖和细胞死亡的影响。

结果
