Abstract

OBJECTIVE

To study the process in vivo of the components in the precipitation of Xiexin decoction.

METHOD

The components in the rats' urine samples after oral administration of baicalin, single herb decoction of Radix Scutellariae or the precipitation of Xiexin decoction were analyzed by LC-MS-MS method; and the absorption of baicalin in different samples were calculated by intestinal in situ loop method.

RESULT

The urinary excretion amounts of baicalin in three samples were not obviously different, whereas the time reach elimination-peak of baicalin in them had significant difference. The absorption of baicalin in the precipitation was obviously greater than that in the single herb decoction and single baicalin. We found that wogonoside and 7-O-glucuronide chrysin were the metabolites presenting in the rat urine samples after oral administration of baicalin by LC-MS-MS, which had never been reported.

CONCLUSION

The resident time in the body of baicalin in the precipitation is prolonged, compared with that in the single herb decoction and single baicalin. The metabolites may be the potential biological components in vivo of baicalin in the precipitation.

目的

研究泻心汤沉淀中各成分的体内过程。

方法

采用液相色谱-串联质谱法分析大鼠口服黄芩苷、黄芩单味药煎剂或泻心汤沉淀后尿样中的成分;采用肠原位灌流法计算不同样品中黄芩苷的吸收情况。

结果

三个样品中黄芩苷的尿排泄量无明显差异,但黄芩苷在其中达到消除峰的时间有显著差异。沉淀中黄芩苷的吸收明显大于单味药煎剂和单纯黄芩苷。通过液相色谱-串联质谱法发现,黄芩苷口服给药后大鼠尿样中出现的代谢产物为汉黄芩苷和7-O-葡萄糖醛酸基白杨素,此前未见报道。

结论

与单味药煎剂和单纯黄芩苷相比,沉淀中黄芩苷在体内的驻留时间延长。这些代谢产物可能是沉淀中黄芩苷在体内的潜在生物活性成分。
