To establish a method of constructing skin-equivalents (SE) by the hair follicle stem cells (HFSC) and the fibroblasts.
The K19 immunostaining was employed to localize the HFSC in the human scalp from the cosmetic surgery. The isolated HFSC through the enzyme digestion were seeded on the dermal equivalent (DE) formed by polymerization of the fibroblasts and collagen. After being cultured between the air-liquid interface for 14 days, SE were harvested and used for an evaluation.
HFSC were located mainly in the outer root sheath in the hair follicle. Based on DE, the growing HFSC could build a fully-developed and multi-layered epidermis with the basal membrane formed between the epidermis and the dermis. The fibroblasts were active and spread evenly in the collagen matrix.
The hair follicle stem cells located in the outer root sheath can be successfully used to construct skin-equivalents in vitro and have a promising clinical use in the treatment.
建立一种利用毛囊干细胞(HFSC)和成纤维细胞构建皮肤替代物(SE)的方法。
采用K19免疫染色法对整形手术获取的人头皮毛囊干细胞进行定位。通过酶消化分离得到的毛囊干细胞接种于成纤维细胞与胶原蛋白聚合形成的真皮替代物(DE)上。在气液界面培养14天后,收获皮肤替代物并进行评估。
毛囊干细胞主要位于毛囊外根鞘。基于真皮替代物,生长的毛囊干细胞可构建出发育完全且多层的表皮,并在表皮与真皮之间形成基底膜。成纤维细胞活性良好且在胶原基质中均匀分布。
位于外根鞘的毛囊干细胞可成功用于体外构建皮肤替代物,在治疗方面具有广阔的临床应用前景。
