Abstract

AIM

To study the effect of Arg-Gly-Asp-Ser (RGDS) tetrapeptide on the expression of matrix metalloproteinase-13 (MMP-13) and the tissue inhibitor of metalloproteinase-1(TIMP-1) in hepatic stellate cell(HSC) stimulated by fibronectin (FN).

METHODS

The expression of MMP-13 was evaluated by RT-PCR. TIMP-1 was analyzed by in-situ hybridization at mRNA level and by Western blot at protein level.

RESULTS

Treated by RGDS tetrapeptide for 2 h, the mRNA expression of MMP-13 was upregulated but the expression of TIMP-1 mRNA and its protein in HSC was inhibited.

CONCLUSION

The expression of MMP-13 can be induced by RGDS tetrapeptide and the expression of TIMP-1 can be inhibited by RGDS tetrapeptide, which is one of the mechanisms of its anti-fibrogenesis.

目的

研究精氨酸-甘氨酸-天冬氨酸-丝氨酸(RGDS)四肽对纤连蛋白(FN)刺激的肝星状细胞(HSC)中基质金属蛋白酶-13(MMP-13)和金属蛋白酶组织抑制剂-1(TIMP-1)表达的影响。

方法

采用逆转录-聚合酶链反应(RT-PCR)评估MMP-13的表达。通过原位杂交在mRNA水平分析TIMP-1,并在蛋白质水平通过蛋白质免疫印迹法进行分析。

结果

用RGDS四肽处理2小时后,HSC中MMP-13的mRNA表达上调,但TIMP-1 mRNA及其蛋白的表达受到抑制。

结论

RGDS四肽可诱导MMP-13的表达,抑制TIMP-1的表达,这是其抗纤维化作用机制之一。
