To test the effect of lycopene on the proliferation and apoptosis of the estrogen receptor(ER)-positive MCF-7 and ER-negative MDA-MB-231 human breast cancer cell lines.
The cell proliferation was analyzed by the MTT and the H3-TdR incorporation. The effect of lycopene on the cell cycle and apoptosis of the synchronized cells was observed through flow cytometry.
Lycopene inhibited the growth and DNA synthesis of both ER-positive MCF-7 and ER-negative MDA-MB-231 cells in a dose-dependent pattern. The maximum inhibition effect appeared after 4 days of exposure to lycopene, with an inhibition rate of 52.6% and 61.9% for the MCF-7 and MDA-MB-231 cells respectively. The flow cytometry analysis found more cells in the G0/G1 phase and less cells in the S and G2/M phase after 24 hours of exposure to lycopene. Lycopene induced apoptosis for the MDA-MB-231 cells, but not for the MCF-7 cells.
Lycopene inhibits the growth of ER-positive MCF-7 cells through the inhibition of the cell cycle progression. The inhibition of ER- negative MDA-MB-231 cells by lycopene is associated not only with the G1 phase cell cycle-arrest but also the induction of apoptosis.
检测番茄红素对雌激素受体(ER)阳性的MCF-7和ER阴性的MDA-MB-231人乳腺癌细胞系增殖和凋亡的影响。
采用MTT法和³H-TdR掺入法分析细胞增殖情况。通过流式细胞术观察番茄红素对同步化细胞的细胞周期和凋亡的影响。
番茄红素以剂量依赖性方式抑制ER阳性的MCF-7细胞和ER阴性的MDA-MB-231细胞的生长及DNA合成。暴露于番茄红素4天后出现最大抑制效应,MCF-7细胞和MDA-MB-231细胞的抑制率分别为52.6%和61.9%。流式细胞术分析发现,暴露于番茄红素24小时后,更多细胞处于G0/G1期,而处于S期和G2/M期的细胞减少。番茄红素可诱导MDA-MB-231细胞凋亡,但对MCF-7细胞无此作用。
番茄红素通过抑制细胞周期进程抑制ER阳性的MCF-7细胞生长。番茄红素对ER阴性的MDA-MB-231细胞的抑制作用不仅与G1期细胞周期阻滞有关,还与诱导凋亡有关。
