To detect the expression of proliferating cell nuclear antigen (PCNA) in severely damaged intestinal mucosa due to high-dose 5-FU exposure.
Thirty-two adult C57BL/6J mice were subjected to daily intraperitoneal high-dose 5-FU injection at 150 mg/kg for 5 consecutive days, and on days 1, 3, and 5, the mice were sacrificed to obtain the small intestinal tissue for HE straining and immunohistochemistry for detecting PCNA expression. Another 8 mice with intraperitoneal PBS injection served as the control group.
High-dose 5-FU exposure of the mice resulted in severe intestinal mucous damage, with complete destruction of the villi and crypts and significantly increased cells positive for PCNA expression (P<0.01).
High-dose 5-FU treatment can significantly increase the PCNA index, and the cells expressing PCNA can be closely associated with regeneration of the severely damaged mucosa due to the exposure.
检测高剂量5-氟尿嘧啶(5-FU)暴露所致严重受损肠黏膜中增殖细胞核抗原(PCNA)的表达。
32只成年C57BL/6J小鼠连续5天每天腹腔注射150mg/kg高剂量5-FU,于第1、3和5天处死小鼠,获取小肠组织进行HE染色和免疫组织化学检测PCNA表达。另外8只腹腔注射PBS的小鼠作为对照组。
小鼠高剂量5-FU暴露导致严重的肠黏膜损伤,绒毛和隐窝完全破坏,PCNA表达阳性细胞显著增加(P<0.01)。
高剂量5-FU处理可显著增加PCNA指数,且PCNA表达细胞可能与该暴露所致严重受损黏膜的再生密切相关。
