To detect aquaporin1 (AQP1) expression in normal human placenta and fetal membranes.
Human placenta and fetal membrane specimens were collected from 5 pregnant women with intact membranes undergoing elective cesarean sections at term. Expression and localization of AQP1 was detected by RT-PCR, Western blotting and immunohistochemistry.
AQP1 mRNA was detected in the placenta and fetal membranes by RT-PCR, and Western blotting also yielded positive results for the specimens, showing a specific band around 28 kD. Immunohistochemical staining showed AQP1 expression in the vascular endothelial cells and syncytiotrophoblasts of the placenta, epithelial cells of the amnion, and cytotrophoblasts of the chorion.
The presence of AQP1 expression in the placenta and fetal membranes suggest that AQP1 may play an important role in maternal-fetal fluid exchange and amniotic fluid balance.
检测水通道蛋白1(AQP1)在正常人类胎盘和胎膜中的表达。
从5例足月行择期剖宫产且胎膜完整的孕妇中收集人胎盘和胎膜标本。采用逆转录聚合酶链反应(RT-PCR)、蛋白质免疫印迹法和免疫组织化学法检测AQP1的表达及定位。
RT-PCR检测显示胎盘和胎膜中有AQP1 mRNA,蛋白质免疫印迹法对标本检测也呈阳性结果,显示在约28 kD处有一条特异性条带。免疫组织化学染色显示AQP1在胎盘的血管内皮细胞和合胞体滋养层细胞、羊膜上皮细胞以及绒毛膜的细胞滋养层细胞中表达。
胎盘和胎膜中存在AQP1表达提示AQP1可能在母胎液体交换和羊水平衡中起重要作用。
