Abstract

OBJECTIVE

To study the localization of ferroportin1 (FP1) protein, the protein and mRNA expression of FP1 in the last trimester placenta of different iron status of pregnant women.

METHODS

Forty placentas were collected from term pregnant women with different degrees of iron deficiency. The localization of FP1 protein of the placenta was determined by immunohistochemical staining. The expressions of FP1 protein and mRNA were assessed by means of Western Blot and real-time quantitative RT-PCR.

RESULTS

The results showed that FP1 protein was localized in the placental syncytiotrophoblast (STB) cells and predominantly in the cytoplasm. There were no significant changes at both protein and mRNA expression levels of placental FP1 of the pregnant women with different iron status.

CONCLUSION

The expression of both protein and mRNA of FP1 of term placentas was not influenced by different degree of iron status of pregnant women.

目的

研究铁转运蛋白1(FP1)在不同铁状态孕妇晚期胎盘组织中的定位,以及FP1蛋白和mRNA的表达情况。

方法

收集40例不同程度缺铁的足月孕妇胎盘组织。采用免疫组化染色法检测胎盘组织中FP1蛋白的定位。采用蛋白质免疫印迹法(Western Blot)和实时定量逆转录聚合酶链反应(real-time quantitative RT-PCR)检测FP1蛋白和mRNA的表达。

结果

结果显示,FP1蛋白定位于胎盘合体滋养层(STB)细胞,且主要位于细胞质中。不同铁状态孕妇胎盘组织中FP1的蛋白和mRNA表达水平均无显著变化。

结论

足月胎盘组织中FP1的蛋白和mRNA表达不受孕妇不同程度铁状态的影响。
