To study the correlation of inhibitor of differentiation 1 (Id-1) to tumor invasion and metastasis by examining Id-1 expression levels in different stages of cervical carcinogenesis.
Id-1 mRNA and protein expression was detected in total of 171 cervical samples including precancerous and cancerous tissues by quantitative RT-PCR (qRT-PCR) and immunohistochemistry (IHC), respectively. Twenty-five normal cervical tissues were used as a normal control. Correlation between Id-1 positive rates and expression levels to cancer progression and clinicopathologic features was statistically analyzed.
A gradual increase of Id-1 protein expression associated with cervical cancer progression was detected (4%, 16%, 50% and 75.9% in normal, low squamous intraepithelial lesion (LSIL), high squamous intraepithelial lesion (HSIL) and cancer tissue, respectively, p<0.001). A similar trend of Id-1 mRNA expression was also observed (1.3, 3.4 and 10.4 fold higher than normal tissues in LSIL, HSIL and cancer tissue, respectively, p<0.001). Furthermore, the Id-1 expression level was correlated to tumor grade (p=0.005), lymph node metastasis (p=0.001), interstitial invasive (p<0.001) and tumor size (p<0.001). These results suggest that high Id-1 expression is associated with tumor growth, invasion and metastasis.
Id-1 expression is correlated to progression and aggressive behaviors in cervical cancer, suggesting a tumor-promoting role for Id-1 in progression of this malignancy.
通过检测分化抑制因子1(Id-1)在宫颈癌发生不同阶段的表达水平,研究其与肿瘤侵袭和转移的相关性。
分别采用定量逆转录聚合酶链反应(qRT-PCR)和免疫组织化学(IHC)检测171例宫颈样本(包括癌前组织和癌组织)中Id-1 mRNA和蛋白的表达。25例正常宫颈组织用作正常对照。对Id-1阳性率和表达水平与癌症进展及临床病理特征之间的相关性进行统计学分析。
检测到Id-1蛋白表达随宫颈癌进展逐渐升高(正常、低级别鳞状上皮内病变(LSIL)、高级别鳞状上皮内病变(HSIL)和癌组织中的表达率分别为4%、16%、50%和75.9%,p<0.001)。Id-1 mRNA表达也观察到类似趋势(LSIL、HSIL和癌组织中的表达分别比正常组织高1.3、3.4和10.4倍,p<0.001)。此外,Id-1表达水平与肿瘤分级(p=0.005)、淋巴结转移(p=0.001)、间质浸润(p<0.001)和肿瘤大小(p<0.001)相关。这些结果表明,Id-1高表达与肿瘤生长、侵袭和转移相关。
Id-1表达与宫颈癌的进展和侵袭性行为相关,提示Id-1在该恶性肿瘤进展中具有促肿瘤作用。
