The intracellular antioxidant system, based on glutathione (GSH), plays a key role in endometrial detoxification reactions and has been proposed to be involved in the pathogenesis endometriosis. This study was designed to evaluate whether estradiol (E(2)) and proinflammatory cytokines have any effects on expression of glutathione in endometrial stromal cells (ESCs).
Glutathione levels were measured utilizing high-performance liquid chromatography following in vitro culture and treatment of ESCs with estradiol, tumor necrosis factor-alpha (TNF-alpha) and interleukin 1-beta (IL-1beta).
The GSH level in E(2) (10(-8) m) treatment group was significantly higher than in the control group at 48 h (P < 0.05). In vitro treatment of ESCs with TNF-alpha 10 ng/mL as well as E(2) (10(-8) m) plus TNF-alpha 10 ng/mL for 48 hr also led to a significant increase in GSH level (P < 0.05; P < 0.05, respectively). Both IL-1beta 10 ng/mL and E(2) (10(-8) m) plus IL-1beta 10 ng/mL for 48 hr increased GSH level significantly (P < 0.05; P < 0.05, respectively) as well.
These findings might suggest that increased production of estradiol and proinflammatory cytokines in the peritoneal cavity possibly leads to the establishment of endometriosis through increased level of GSH.
基于谷胱甘肽(GSH)的细胞内抗氧化系统在子宫内膜解毒反应中发挥关键作用,并且据推测其与子宫内膜异位症的发病机制有关。本研究旨在评估雌二醇(E2)和促炎细胞因子是否对子宫内膜基质细胞(ESCs)中谷胱甘肽的表达有任何影响。
采用高效液相色谱法测定体外培养和用雌二醇、肿瘤坏死因子-α(TNF-α)和白细胞介素 1-β(IL-1β)处理 ESCs 后 GSH 水平。
E2(10-8 m)处理组的 GSH 水平在 48 小时时明显高于对照组(P<0.05)。体外用 TNF-α 10ng/mL 以及 E2(10-8 m)加 TNF-α 10ng/mL 处理 48 小时也导致 GSH 水平显著升高(P<0.05;P<0.05,分别)。IL-1β 10ng/mL 和 E2(10-8 m)加 IL-1β 10ng/mL 处理 48 小时均显著升高 GSH 水平(P<0.05;P<0.05,分别)。
这些发现可能表明,腹腔内雌二醇和促炎细胞因子的产生增加可能通过增加 GSH 水平导致子宫内膜异位症的发生。
