The alpha subunit of Escherichia coli DNA-dependent RNA polymerase is encoded by the rpoA gene and plays a major role in enzyme assembly. A set of C-terminal deletion mutations of the rpoA gene was constructed. The results of mixed reconstitution experiments in vitro, using the truncated alpha polypeptides encoded by the rpoA deletion mutants, suggest that the amino-terminal two-thirds of alpha subunit is sufficient for the formation of pseudo-core complexes containing both beta and beta' subunits.
大肠杆菌DNA依赖性RNA聚合酶的α亚基由rpoA基因编码,在酶组装中起主要作用。构建了一组rpoA基因的C末端缺失突变体。使用rpoA缺失突变体编码的截短α多肽进行体外混合重组实验的结果表明,α亚基的氨基末端三分之二足以形成包含β和β'亚基的假核心复合物。
