To observe the expression of CD4(+) CD25(+) Foxp3(+) Treg, IL-10 and TGF-beta in lymph and blood, as well as the effect of dexamethasone on it and investigate the relationship between them.
Lymph and blood samples of 0 h, 24 h, 48 h after the last challenge were collected from the rat model of asthma. The percentage of CD4(+) CD25(+) Foxp3(+) Treg were detected by flow cytometer (FCM), while the levels of IL-10 and TGF-beta were determined by ELISA.
The percentage of CD4(+) CD25(+) Foxp3(+) Treg in the blood and the levels of IL-10 and TGF-beta in the plasma in cases of asthma group were significantly lower than that of the control and therapy group at different time points (P<0.05); The percentage of CD4(+) CD25(+) Foxp3(+) Treg and the level of IL-10 in lymph were significantly higher than that of blood (P<0.05), however, the level of TGF-beta in lymph were significantly lower than that of blood (P<0.05); There were no significant differences which the percentage of CD4(+) CD25(+) Foxp3(+) Treg in the blood and the levels of IL-10 and TGF-beta in the plasma between control group and therapy group (P>0.05).
There is a disbalance both quantity and function of CD4(+) CD25(+) Foxp3(+) in the lymph of bronchial asthmatic rat, and the percentage of CD4(+) CD25(+) Foxp3(+) Treg in lymph are significantly higher than that of blood, which dexamethasone may effectively increase the percentage of CD4(+) CD25(+) Foxp3(+) Treg in bronchial asthmatic rat.
观察哮喘大鼠模型末次激发后0 h、24 h、48 h 时其淋巴液和血液中CD4(+)CD25(+)Foxp3(+)调节性T细胞(Treg)、白细胞介素-10(IL-10)及转化生长因子-β(TGF-β)的表达情况,以及地塞米松对其的影响,并探讨它们之间的关系。
采集哮喘大鼠模型末次激发后0 h、24 h、48 h的淋巴液和血液样本。采用流式细胞仪(FCM)检测CD4(+)CD25(+)Foxp3(+)Treg的百分比,采用酶联免疫吸附测定(ELISA)法检测IL-10和TGF-β的水平。
哮喘组不同时间点血液中CD4(+)CD25(+)Foxp3(+)Treg的百分比及血浆中IL-10和TGF-β的水平均显著低于对照组和治疗组(P<0.05);淋巴液中CD4(+)CD25(+)Foxp3(+)Treg的百分比及IL-10水平均显著高于血液(P<0.05),而淋巴液中TGF-β水平显著低于血液(P<0.05);对照组和治疗组血液中CD4(+)CD25(+)Foxp3(+)Treg的百分比及血浆中IL-10和TGF-β的水平比较,差异无统计学意义(P>0.05)。
支气管哮喘大鼠淋巴液中CD4(+)CD25(+)Foxp3(+)Treg在数量和功能上均存在失衡,且淋巴液中CD4(+)CD25(+)Foxp3(+)Treg的百分比显著高于血液,地塞米松可有效提高支气管哮喘大鼠CD4(+)CD25(+)Foxp3(+)Treg的百分比。
