To explore the expression level of insulin-like growth factor-1 receptor (IGF-IR) in malignant clone cells of myelodysplastic syndromes (MDS).
Fluorescence in situ hybridization (FISH) and immunochemistry (alkaline phosphatase antialkaline phosphatase) were used together to detect the expression of IGF-IR in the bone marrow cells of 26 MDS patients with known abnormal karyotypes.
The average IGF-IR expression level on the surface of clone cells from the 26 MDS cases was markedly elevated compared with the corresponding level in normal cells (78.2 ± 13.7% vs. 14.1 ± 14.0%, p < .0001). The percentages of malignant clone cells in all 26 MDS cases were significantly correlated with the respective percentages of IGF-IR-positive nucleated cells (r = 0.909; p < .0001). No significant difference in the IGF-IR expression level on the clone cells were observed either between high- and low-risk MDS patients or among MDS patients with different abnormal karyotypes.
IGF-IR might be taken as a marker of clone cells in MDS.
探讨骨髓增生异常综合征(MDS)恶性克隆细胞中胰岛素样生长因子-1 受体(IGF-1R)的表达水平。
采用荧光原位杂交(FISH)和免疫组织化学(碱性磷酸酶抗碱性磷酸酶)联合检测 26 例已知染色体异常的 MDS 患者骨髓细胞中 IGF-1R 的表达。
26 例 MDS 患者克隆细胞表面的 IGF-1R 表达水平平均明显高于相应正常细胞(78.2±13.7%比 14.1±14.0%,p<0.0001)。26 例 MDS 患者中所有恶性克隆细胞的比例与各自 IGF-1R 阳性核细胞的比例呈显著相关(r=0.909;p<0.0001)。高风险和低风险 MDS 患者之间以及不同染色体异常 MDS 患者之间克隆细胞的 IGF-1R 表达水平无显著差异。
IGF-1R 可作为 MDS 克隆细胞的标志物。
