It was proven that Vitamin C could inhibit the growth of many types of tumors as an antioxidant. The aim of this study is to explore role of Vitamin C in proliferation and apoptosis of lung carcinoma cell line A549 and the underlying mechanism.
A549 cells were cultured in vitro and incubated with Vitamin C. The cell viability was measured by growth curve and clonogentic assay. Flow cytometry was used to analyze cell cycle and detect apoptosis. The levels of expression of Caspase-3 mRNA and Survivin mRNA were detected by RT-PCR.
Vitamin C of 400 microg/mL, 4 mg/mL significantly inhibited the growth of A549 cell lines (P = 0.024, P = 0.015, respectively). Flow cytometry showed that the cells major stagnation stayed in the G0/G1 and S phase and the apoptotic rate increased with time prolonged. Vitamin C signifiantly up-regulated the expression of Caspase-3 mRNA, but had no effect on Survivin mRNA.
Vitamin C can inhibit the proliferation of A549, block A549 cells in G0/G1 and S phase, and induce apoptosis of A549 cells. Apotosis occurred by up-regulated the expression of Caspase-3.

体外培养A549细胞,并与维生素C孵育。通过生长曲线和克隆形成试验检测细胞活力。采用流式细胞术分析细胞周期并检测细胞凋亡。通过逆转录-聚合酶链反应(RT-PCR)检测半胱天冬酶-3(Caspase-3)mRNA和生存素(Survivin)mRNA的表达水平。
400μg/mL、4mg/mL的维生素C显著抑制A549细胞系的生长(分别为P = 0.024,P = 0.015)。流式细胞术显示,细胞主要停滞在G0/G1期和S期,且凋亡率随时间延长而增加。维生素C显著上调Caspase-3 mRNA的表达,但对Survivin mRNA无影响。
维生素C可抑制A549细胞的增殖,使A549细胞阻滞于G0/G1期和S期,并诱导A549细胞凋亡。凋亡通过上调Caspase-3的表达而发生。
