Abstract
FoxO1 regulates the transcription of genes involved in diabetes, cancer and ageing. In this issue of , Olefsky and colleagues report that FoxO1 also causes inflammation by regulating the pro-inflammatory Tlr4 signalling pathway in macrophages. Insulin-mediated activation of Akt inactivates FoxO1 and thereby limits the inflammatory response.
