A method of stereotaxic apparatus was employed to damage the nucleus basalis of Meynert, and the effects of Tiao-xinzishen prescriptions on neurotransmitters such as ACh, 5-HT and NE in hippocampus were observed.
Rats were placed on the brain stereotaxic apparatus and received a bilateral lesion of Meynert by IA injection, according to the atlas of Daxinos and Watson. After seven days of lesion, AD rats were selected. Rats were treated with Tiaoxin or/and Zishen prescription for 20 days, respectively. Hippocampus ACh was measured by spectrophotometer and 5-HT and NE by high performance liquid chromatography (HPLC).
The contents of hippocampus ACh, 5-HT and NE of AD rats were significantly increased after the treatment with the three prescriptions, respectively.
These prescriptions had some up regulating effects on hippocampus neurotransmitters in rats, which had already decreased due to dementia.
采用立体定向仪损毁Meynert基底核,观察调心滋肾方对海马区乙酰胆碱(ACh)、5-羟色胺(5-HT)和去甲肾上腺素(NE)等神经递质的影响。
根据Daxinos和Watson图谱,将大鼠置于脑立体定向仪上,通过脑室内注射对Meynert核进行双侧损毁。损毁7天后,选取阿尔茨海默病(AD)大鼠。大鼠分别用调心方或/和滋肾方治疗20天。用分光光度计测定海马区ACh含量,用高效液相色谱法(HPLC)测定5-HT和NE含量。
三种方剂治疗后,AD大鼠海马区ACh、5-HT和NE含量均显著升高。
这些方剂对因痴呆而降低的大鼠海马区神经递质有一定的上调作用。
