To observe the effect of acupotomy lysis (AL) on hypothalamic proopiomelanocortin (POMO) mRNA and preproenkephalin (PPE) mRNA expression in rats with the third lumbar vertebrae transverse process syndrome (TLVTPS) so as to study its underlying mechanism in relieving symptoms of lumbar muscle strain.
Twenty-four SD rats were randomly divided into normal control group, model group, AL group and electroacupunture (EA) group. The TLVTPS model was established by inserting a piece of gelatin sponge into the posterior of the left 3rd lumbar vertebrae transverse process. AL and EA were applied to the left "Shenshu"(BL 23) and "Yaoyangguan" (GV 3) respectively. The POMC mRNA and PPE mRNA expression levels in the hypothalamus were detected by in situ hybridization.
In comparison with the normal group, the integrated optical density (IOD) values of hypothalamic POMC mRNA and PPE mRNA positive cells in the model group were increased significantly (P < 0.01); while compared with the model group, those of POMC mRNA and PPE mRNA positive cells in both left and right hypothalamus were increased further considerably in both AL and EA groups (P < 0.01). No significant differences were found between AL and EA groups in POMC mRNA and PPE mRNA expression levels (P > 0.05).
AL and EA therapies can increase the expression of POMC mRNA and PPE mRNA in hypothalamus in rats with TLVTPS, which may contribute to its effect in relieving pain in the treatment of lumbar muscle strain.
观察针刀松解术(AL)对第三腰椎横突综合征(TLVTPS)大鼠下丘脑阿片促黑素皮质素原(POMO)mRNA和前脑啡肽原(PPE)mRNA表达的影响,以探讨其缓解腰肌劳损症状的潜在机制。
将24只SD大鼠随机分为正常对照组、模型组、针刀松解组和电针组。通过将一片明胶海绵插入左侧第三腰椎横突后方建立TLVTPS模型。分别对左侧“肾俞”(BL 23)和“腰阳关”(GV 3)进行针刀松解术和电针治疗。采用原位杂交法检测下丘脑POMC mRNA和PPE mRNA的表达水平。
与正常组相比,模型组下丘脑POMC mRNA和PPE mRNA阳性细胞的积分光密度(IOD)值显著升高(P < 0.01);与模型组相比,针刀松解组和电针组双侧下丘脑POMC mRNA和PPE mRNA阳性细胞的IOD值均进一步显著升高(P < 0.01)。针刀松解组和电针组在POMC mRNA和PPE mRNA表达水平上无显著差异(P > 0.05)。
针刀松解术和电针疗法均可增加TLVTPS大鼠下丘脑POMC mRNA和PPE mRNA的表达,这可能是其在治疗腰肌劳损中缓解疼痛的作用机制。
