To explore protective effect of probucol on liver injury induced by carbon tetrachloride (CCl4) in rats.
We used CCl4 to induce a rat model of liver injury. Some of those rats were treated with an oral administration of probucol. Liver function, blood lipid, liver tissue malondialdehyde (MDA), superoxide dismutase (SOD), and degree of liver fibrosis were analyzed.
Probucol significantly decreased the elevation of serum aspartate transaminase, total bile acid, alkaline phosphatase and cholesterol, and improved liver histopathological changes (including fatty infiltration, inflammation cell infiltration, and fibrosis), decreased liver tissue MDA level, and increased liver tissue SOD level in liver injury rats.
For the first time, we demonstrated that probucol has protective effect against liver injury in animal experiment. The antioxidant action of probucol may play an important role in its hepatoprotective mechanism. Probucol also can reduce serum cholesterol. Thus, probucol may have the potential use in clinical liver diseases in which oxidative stress may be present, especially for the patients with hypercholesterolemia.
探讨普罗布考对四氯化碳(CCl4)诱导的大鼠肝损伤的保护作用。
我们用CCl4诱导大鼠肝损伤模型。其中一些大鼠口服普罗布考进行治疗。分析肝功能、血脂、肝组织丙二醛(MDA)、超氧化物歧化酶(SOD)及肝纤维化程度。
普罗布考显著降低肝损伤大鼠血清天冬氨酸转氨酶、总胆汁酸、碱性磷酸酶及胆固醇的升高,改善肝脏组织病理学变化(包括脂肪浸润、炎症细胞浸润及纤维化),降低肝组织MDA水平,升高肝组织SOD水平。
我们首次在动物实验中证明普罗布考对肝损伤具有保护作用。普罗布考抗氧化作用可能在其肝保护机制中起重要作用。普罗布考还可降低血清胆固醇。因此,普罗布考可能对存在氧化应激的临床肝脏疾病具有潜在应用价值,尤其是对高胆固醇血症患者。
