Abstract

OBJECTIVE

To determine the relationship of thick greasy tongue fur formation and permeability of vascular endothelial cells (ECs) with the protein expression of zonula occludens-1 (ZO-1).

METHODS

Sprague Dawley rats were randomly divided into a model group of severe acute pancreatitis (SAP) and a sham-operated (SO) group. The SAP rats were further divided into two subgroups on the basis of tongue-coating status: a thick greasy tongue fur group (SAP-TGF) and a normal tongue fur group (SAP-NF). Six rats were chosen randomly from every group mentioned above for an Evans blue assay 5 days after model establishment. For the histomorphology analysis, the expressions of ZO-1 protein and mRNA were studied by hematoxylin-eosin (H&E) staining, transmission electron microscope, Western blot, and Q-PCR using blood and tongue tissues, which were collected from 8 rats randomly chosen from each group.

RESULTS

The papillae density of the rat tongue surface and the caryocinesis frequency of the basal layer were significantly increased in the SAP-TGF group compared with the SO group (P<0.05). Evans blue levels in the tongue tissue of the SAP-TGF group were significantly higher than that of the SO and SAP-NF groups (P<0.05). Vascular ECs were wider and obviously fissured in the SAP-TGF group under transmission electron microscope observation. The protein and mRNA expression of ZO-1 in the SAP-TGF group were lower than those in the SAP-NF (P<0.05).

CONCLUSIONS

Reproductive activity enhancement of glossal epithelial cells was one of the main characteristics of thick greasy tongue fur formation. An increase in vasopermeability was closely associated with thick greasy tongue fur formation. Tight junction structural variation of vascular ECs might play an important role in the pathological and physiological process of thick greasy tongue fur formation.

探讨厚腻苔形成与血管内皮细胞(endothelial cells,ECs)通透性的关系及其与紧密连接蛋白-1(zonula occludens-1,ZO-1)表达的关系。

方法

将 Sprague Dawley 大鼠随机分为重症急性胰腺炎(severe acute pancreatitis,SAP)模型组和假手术(sham operated,SO)组,SAP 大鼠再根据舌苔情况分为厚腻苔组(SAP-TGF)和薄白苔组(SAP-NF),每组设 6 只大鼠,建模后第 5 天采用伊文思蓝(Evans blue,EB)法检测大鼠舌组织通透性;每组随机取 8 只大鼠,采用苏木精-伊红(hematoxylin-eosin,HE)染色、透射电镜、Western blot 和实时荧光定量 PCR(quantitative real-time PCR,Q-PCR)检测各组大鼠舌组织和血液中 ZO-1 蛋白及 mRNA 的表达。

结果

与 SO 组比较,SAP-TGF 组大鼠舌乳头密度和基底层细胞有丝分裂频率显著增加(P<0.05);SAP-TGF 组大鼠舌组织 EB 含量明显高于 SO 组和 SAP-NF 组(P<0.05);透射电镜下观察到 SAP-TGF 组大鼠血管内皮细胞(endothelial cells,ECs)明显增宽,出现裂隙;SAP-TGF 组大鼠舌组织和血液中 ZO-1 蛋白及 mRNA 的表达均明显低于 SAP-NF 组(P<0.05)。

结论

厚腻苔形成的主要特征之一是舌黏膜上皮细胞的有丝分裂活跃,血管通透性增加与厚腻苔形成密切相关,血管 ECs 紧密连接结构的改变可能在厚腻苔形成的病理生理过程中发挥重要作用。
