To study the anti-tumor metastatic constituents from Ardisia Crenata.
Chemical constituents were isolated and purified by repeated column chromatography( silica gel, Toyopearl HW40C and preparative HPLC). Their structures were elucidated on the basis of spectral data analysis. The anti-tumor metastasis assay was applied to evaluate the isolated compounds of their activities.
Nine compounds(1-9) were isolated and their structures were identified by comparison of their spectral data with literature values as follows: 5-hydroxymethyl-2-furalclehyde(1), ethyl-beta-D-fructopyranoside(2), syringic acid(3), n-butyl-beta-D-fructofuranoside(4), n-butyl-alpha-D-fructofuranoside(5), methyl-alpha-D-fructofuranoside(6), (+)-bergenin(7), ardisiacrispins B(8), asperuloside acid(9). The isolated compounds(1-9) showed positive anti-tumor metastatic activities, and compounds 1, 5, and 8 showed significant anti-tumor metastatic activities. At the concentration of 0.8 mg x L(-1), compound 5 revealed the value of metastatic inhibition ratio on MDA-MB-231 was 93.8%.
Compounds 2-6 and 9 were isolated from this plant for the first time. compounds 1, 5 and 8 showed significant anti-tumor metastatic activities.
研究朱砂根中的抗肿瘤转移成分。
通过反复柱色谱法(硅胶、Toyopearl HW40C和制备型高效液相色谱)分离纯化化学成分。基于光谱数据分析阐明其结构。应用抗肿瘤转移试验评估分离得到的化合物的活性。
分离得到9个化合物(1 - 9),通过将其光谱数据与文献值比较鉴定其结构如下:5-羟甲基-2-糠醛(1)、β-D-果糖吡喃糖苷乙酯(2)、丁香酸(3)、β-D-呋喃果糖苷正丁酯(4)、α-D-呋喃果糖苷正丁酯(5)、α-D-呋喃果糖苷甲酯(6)、(+)-岩白菜素(7)、朱砂根素B(8)、车叶草苷酸(9)。分离得到的化合物(1 - 9)显示出阳性抗肿瘤转移活性,化合物1、5和8显示出显著的抗肿瘤转移活性。在浓度为0.8 mg·L⁻¹时,化合物5对MDA-MB-231的转移抑制率为93.8%。
化合物2 - 6和9首次从该植物中分离得到。化合物1、5和8显示出显著的抗肿瘤转移活性。
