Abstract

OBJECTIVE

To investigate the apoptosis-inducing effect of Ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid (5F), a compound isolated from Pteris semipinnata L (PsL), on human gastric cancer SGC7901 cells and explore its mechanism.

METHODS

The inhibitory effect of 5F on SGC7901 cells was observed by MTT assay and flow cytometry, and the changes of the expression of Bcl-2 and Bax in SGC7901 cells following 5F exposure were evaluated by Western blot analysis.

RESULTS

5F inhibited the proliferation of SGC7901 cells in a concentration- and time-dependent manner, and the cell apoptosis induced by 5F was confirmed by Annexin V-EGFP staining and caspase-3 activation assay. The cell apoptosis induced by 5F was associated with decreased Bcl-2 and increased Bax expressions.

CONCLUSION

5F exposure induces apoptosis in SGC7901 cells by activating mitochondrial apoptotic pathways.

目的

研究从半边旗(PsL)中分离得到的化合物Ent-11α-羟基-15-氧代-贝壳杉-16-烯-19-酸(5F)对人胃癌SGC7901细胞的诱导凋亡作用并探讨其机制。

方法

采用MTT法和流式细胞术观察5F对SGC7901细胞的抑制作用,通过蛋白质免疫印迹分析评估5F作用后SGC7901细胞中Bcl-2和Bax表达的变化。

结果

5F以浓度和时间依赖性方式抑制SGC7901细胞增殖,Annexin V-EGFP染色和caspase-3激活试验证实5F诱导细胞凋亡。5F诱导的细胞凋亡与Bcl-2表达降低和Bax表达增加有关。

结论

5F通过激活线粒体凋亡途径诱导SGC7901细胞凋亡。
