Abstract

OBJECTIVE

To explore whether Radix Dipsaci (RD) exhibits beneficial effects on tendon healing.

METHODS

An attempt was made to explore the in vitro effects of a hot water extract of RD on gene expression of procollagen Type I (COL1A1), procollagen Type III (COL3A1) and decorin in cultured tendon fibroblasts, and its in vivo effects in a well-established rat model of patellar tendon donor site injury.

RESULTS

It was found that gene expression of COL3A1 and decorin in cultured tendon fibroblasts was significantly increased by RD, but that COL1A1 was not affected. In vivo studies showed that RD increased blood vessels in the wound but did not significantly affect the expression of COL1A1, COL3A1 and decorin at day 14 post-injury. The ultimate tensile stress of the healing tendon was not significantly improved by either local injection or oral administration of hot water extract of RD (P > 0.05).

CONCLUSION

The present findings imply that RD per se does not significantly improve tendon healing. Further investigation of RD in a herbal formula may be necessary to test its efficacy in tendon injuries.

目的

探讨独活是否对肌腱愈合有有益作用。

方法

尝试探讨独活热水提取物对培养的肌腱成纤维细胞中Ⅰ型前胶原(COL1A1)、Ⅲ型前胶原(COL3A1)和饰胶蛋白基因表达的体外作用,及其在已建立的兔髌腱供体部位损伤模型中的体内作用。

结果

发现独活可显著增加培养的肌腱成纤维细胞中 COL3A1 和饰胶蛋白的基因表达,但对 COL1A1 没有影响。体内研究表明,独活增加了伤口中的血管,但在损伤后 14 天,对 COL1A1、COL3A1 和饰胶蛋白的表达没有显著影响。局部注射或口服独活热水提取物均不能显著提高愈合肌腱的最大拉伸应力(P>0.05)。

结论

本研究结果表明,独活本身并不能显著改善肌腱愈合。有必要在草药配方中进一步研究独活,以检验其在肌腱损伤中的疗效。
